Monocytes in umbilical cord blood make up approximately 20% of the total mononucleated cells. CD14+ monocytes play a key role in the immune response, being important to both inflammatory and anti-inflammatory function. Both in vivo and in culture monocytes can differentiate into macrophages, dendritic cells, and mast cells in the presence of the right growth factors. Cord blood derived CD14+ monocytes have a similar cytokine phenotype as their adult derived counterparts. Findings in some fields such as neuroscience and traumatic brain injury have found cord blood monocytes to provide better clinical outcomes. Cord blood monocytes, but not adult derived monocytes, have even been shown to have a therapeutic use in brain injury, accelerating brain remyelination in cell therapy.

Primary human CD14+ cells are isolated from umbilical cord blood (CB) mononuclear cells (MNCs) using positive immunomagnetic separation techniques. CB was collected using citrate-phosphate-dextrose (CPD) as an anticoagulant.
